Fu fatto costruire nel 1580 da Gianfilippo Serlupi su alcune case, appositamente demolite, già di proprietà della sua famiglia sin dal Quattrocento. Il progetto della costruzione fu probabilmente di Giacomo Della Porta, il quale però non vide ultimati i lavori, che terminarono nel 1619 grazie all’intervento del fratello di Gianfilippo, monsignor Girolamo Serlupi.
